Do you mean that you want to prevent the updates on the entire instance or just on one database? If on a database then just remove the update permissions (update, insert, delete) from all users but do store the original permissions first though. If you only want this to happen for a short period of time you could open a transaction and lock all the tables in the database in shared mode. If you want the entire instance to become read-only then either remove all the permissions form all databases as described above or consider bringing up the instance as an HDR secondary which will prevent any updates made on the instance. Although you would have to define an HDR primary and all changes done on the primary instance will be propagated onto the secondary.
