There are several things to monitor, all of which are application-specific. TFM has some general guidelines, in the System Administration manual and the Performance Guide. Check the Informix web site for white papers on this subject, too. The best way to find out about monitoring and tuning is to take the Informix classes on System Administration and Performance Tuning. If you are going to the User Conference this year I expect a presentation on monitoring the OnLine system via onstat and the Sysmaster database.
