I want to make a copy of a 2TB database using cooked files. Most of that space
is sblob. Will it work to copy the data files from the old to the new instance
if my spaces are defined exactly the same on both servers?

Yes, as long as the copy is made while the engine is either offline or
under onmode -c block as if you are taking an external archive.

Thanks Art. My plan is to take a replicated instance offline, ftp the datafiles, then sync everything back up. Saturdays have the lowest transaction load so if I do the copy then I'm hoping syncing things back up afterwards won't be too painful.
Assuming by "replicated instance" you mean an RSS or HDR secondary or an ER replicant. That should work just fine. Art Art S. Just ensure you have enough logical logs so that logs doesn't not get wrapped till you sync up the data to secondary node. Should do, although as per Art the source database, be it the Primary or an RSS/HDR, would have to be offline for the duration of the copy. Have you considered ifxclone for this? It's pretty cool and can be run against a running source.
